In the second half, Lamine Yamal added Spain's third goal in the 63rd minute.
Substitute Nico Williams completed Spain's scoring in the 89th minute.
In the same group, England defeated Czechia by two clean sheets, in the match played at Sinobo Stadium in Czechia.
England's goals were scored by Anthony Gordon in the 47th minute and Harry Kane in the 69th minute.
With this result, Spain took sole possession of the top spot in Group 3 of League A in the UEFA Nations League with 6 points.
England and Croatia are tied on 3 points, but the Three Lions lead in second place due to goal difference, followed by Luka Modric's teammates in third place.
Meanwhile, Czechia sits at the bottom of the group with no points after losing their first two matches.
